How long is Heartbreak Hill in Boston?
Worry about the Newton Hills as a whole, not just Heartbreak. It isn't a big deal standing by itself, but it's placement is what kicks your ass.
Also worry about the big downhill after you get to the top of Heartbreak just as much as you worry about the climb.
It's a relatively flat hill that takes about 1:35 if you are doing hard repeats, but that's not quite the entire hill.
Heartbreak, as a hill, is nothing. Heartbreak at the 20 mile mark of a marathon is something. Be forwarned, once you crest heartbreak there's nothing but screaming downhill ahead -- and that downhill is the hardest part of Boston's course. You can count the number of runners who came through heartbreak in record pace by the dozens, and all of them failed.

Strengthen your quads. Leg extensions, just like Lee Haney tells you to - low weights, high repetitions.
I recommend living in a place with real hills (like CA, CO, etc), where we laugh at the "Newton hills". Man what a total let down those where.
