Will be staying at the Marriott Downtown on Bay St this weekend, looking to get in a short run of 4-6 miles. No car, so must start from hotel. Is it best just to head to the waterfront? Any other options. Thanks.

It's about a mile to the waterfront (Martin Goodman bike path), so that is doable. Probably a better route to head west out-and-back from there.
Also about a mile north to the north half of Queen's Park where the packed gravel path is about half a mile per loop (or pop over to the west for a jaunt through U of T).
There's also a paved bike path down the Rosedale Valley Rd ravine that can give you a bit of an escape from concrete, but has a little longer to get to:

You could also just take the subway up north to Davisville (~15 minutes) and then run point-to-point through Mount Pleasant cemetery and the Moore Park ravine:
Moore Park Ravine is best , but you might get lost . So run this, Go Straight North un Bay Street , turn left on St. Joseph . This will take you to the U of T area , do a loop around campus and head back. 4 miles.
Yes, just go south the mile to the waterfront and then head west. You can go as long as you want as an out-and-back.
If you're only looking for a short run and don't know the city, it would be pretty reasonable to go to Queen's Park and just run the nice half-mile loop there for a while. Very pleasant.
I live downtown and go there all the time. It's especially nice when I need to run at night.
The last time I was in Toronto my hotel had a map of running courses at its front desk.
