Everyone has their opinion but if you are starting out listening to the blues, you should go way back and then move forward to today's artists, many who cover the old songs.
With that being said, you will get a slew of songs suggested as "the greatest blues song ever recorded." Some, like me, would say many of the songs by Robert Johnson, such as Cross Road Blues or Sweet Home Chicago. I would suggest you get 'The Complete Recordings' of RJ but there are many others such as Howlin' Wolf, Son House, Muddy Waters, and Buddy Guy to name a few of my faves.
Also, there are different types of blues such as Delta or Chicago blues, for example.
Do a google search and you will find tons of info. You can even visit the Blues Museum

Like somebody else said, how do you pick the best? I put some of Muddy Waters' recordings at the top of that list...Hoochie Coochie Man, Mannish Boy, and Got My Mojo Working. I think he really captured the real heart and soul of the blues in some those records.
