The BRAIN, my friend. THAT is the most important element.
The Brain is an Organ. Obviously the heart contributes the most during running. In terms of the legs, all the muscles have to work together to allow you to run. Running is not isolated to one muscle like doing curls or something is. If you really have to choose one muscle in the legs, it would probably be the hamstrings because they are the biggest.

you guys are morons. runners typically have strong quads and weak hamstrings. i don't know what muscle contributes the most but it is definately not the hamstrings.
Actually the hamstrings work more then the quads.
The problem sprinters have is that they work too much on
the quad muscles and don't run enough to develop the hamstrings properly.
Distance runners have very defined and strong hamstrings because that is what is worked more while running, comparing to the quad muscles.
Usually the quads will fatigue faster then the hanstrings when distance running.
it would serve you well to visualize your body as one large muscle that is doing it's best to transmit as much energy as possible.
