I work every Thursday, am on the schedule for tomorrow, have never called in sick or not shown up to work, am never late, etc, yet my boss sent me an email after hours to ask me if I was "going to be able to make it tomorrow." What should I make of this?
My boss just emailed me after hours to ask if I would be at work tomorrow when there is no reason why I wouldn't.

You're getting canned and the boss didn't know if HR did it today, or if they are going to do it tomorrow. Asking if you'll be in tomorrow was a subtle way of finding out if they already did the deed.

My manager asks me at least once a week if I'm going to be here tomorrow. As if I'm ever not there.
It's usually because he has a big project for me to start. -

It seems to me the most likely reason is because he plans to bring you a cake tomorrow, or offer you a promotion. If you're really lucky, he's planning to give you a promotion AND a cake!
