The airlines hub and spoke model. After deregulation the major airlines attempted to eliminate competition by concentrating operations and funneling routing/passengers through a central location.
Atlanta has long been a Delta hub and its location is well-suited to control a lot of the air traffic throughout the southeast.
It you consider the 5 busiest airports in the U.S. - ATL, ORD, LAX, DFW, and DEN, all except for LAX are in centrally located cities (relative to the flight network) with large hubs for the major carriers.
By the way, this hub and spoke model also ensures that there are not a lot of unique visitors at any given time. Everyone that flies in the U.S. just keeps getting routed through the same busy airports.

Antarctica probably has the most number of places. Australia probably comes in #2. Gobi is probably pretty high up there.

A couple thoughts -
Delhi, India. One of the biggest cities today and has been inhabited for about 4 millenia (MUCH longer according to some sources, though only ~2600 according to others on the other side of the spectrum).
Karachi, Shanghai and Mumbai are also good guesses... Paris, NYC, Tokyo, Rome and Constantinople are probably other solid guesses.
The single spot where most people have put one foot would be this guy's ass!
The single spot where one human has put their own foot the most times would be this same guy's mouth.
My actual vote would be the main entrance to Grand Central Station. A single spot would see millions of tourists and residents of NYC daily.

Another thing to consider is that larger cities usually have multiple large airports. You might be surprised that Atlanta handles more traffic than, say, JFK just because NYC is so much larger than Atlanta. But NYC has three huge airports. The third largest airport serving the NYC area is bigger than Dulles. Similarly, LA has a few airports other than LAX. So Atlanta isn't a huge city, but all air traffic is constricted to one major airport.

My vote would be the Carmike Grande Movie Theater on Chesnut Street because its real busy every Friday and Saturday. You'd never believe how many people go there, man.
