It's likely that the disk has filesystem corruption. Is it attached to a PC or a Mac? If Mac, then you can check the drive's SMART status, and attempt to fix any filesystem corruption. If Windows, then I have few fvcks to give. In either case, I wouldn't spend too much effort on repair. In fact, repair attempts can make things worse. Did you try rebooting?
Get a second hard drive. If you can get the wonky drive to mount, copy everything immediately. Otherwise, I suggest you buy a copy of Data Rescue. It has saved my bacon more than once.
Once you have everything off the old drive, you can try to reformat it and see if it's good.
