Exactly, the people here saying there is no way are people who either
a)lack athletic ability themselves so 4:30 seems fast
b)have never been around a very high level athlete
There is no way that anyone can run a 4:30 mile without specific training for it.
No NFL players devote the specific training required to do this.
Some may have the ability to do it within one year if they did choose to train specifically for it.
I will use myself as an example.
I played football in high school - wide receiver.
I also sprinted- 11.2, 22.5, 50.2 and a ran a 2:02 800
Then I ran track in college.
My first 1500m race was 2nd year of college and I ran 4:05, which is about a 4:25 mile.
There were a lot of track workouts and distance runs that got me there.
I doubt I could run a 4:30 mile my 1st year of college and I ran 1:55 that year.
I don't see how you run a 4:30 mile with a solid build with football training and I don't see how any football players would be putting in serious distance runs while getting ready for games or even in their off season.
I did get my mile down to 4:04 so I have a wide perspective on different levels of mile fitness.
Another anecdote...
When I was in college one of the guys from the basketball team came out for track his junior year. He ran track his freshman and sophomore years in high school (ran about 4:35-40 or so for the mile), then only ran for general conditioning for basketball until he started track training in college. After two weeks of training with the team he ran a 4:10 1500 in his first meet.
The guy was about 6' 4" and 170 pounds or so (this was a D3 school so it's not like we had giants on our basketball team), but even though he was supremely talented, he was no where near the level of NFL athletes. I realize the conditioning needed for basketball and football are different, but it's not hard to believe that out of the hundreds of NFL players there's at least one with the distance running talent needed to pull off a 4:30.
Some more points not considered in this thread so far:
1) How many miles do you think some of the position players go in practice, especially training camp? On some of the teams that have quick offenses, I'm guessing they go at least 3-4 miles on a daily basis. I know they've put those fit band things on soccer players (some of whom do a 10k in game) - anyone ever seen this done for a football player?
2) I bet there are a few guys that jog on a regular basis at least a few times a week. I'm sure most don't do anything, a bunch ride a stationary bike, so I'm sure there are a few.
3) Fast twitch vs. slow twitch. Yes of course, most football players will be MUCh better suited for 100/200 or even 400m, but there have to be a few that have enough slow twitch and enough miles run (#1 or #2 above) to be able to 4:30.
Summary - I'd say the O/U on this is 2.5 NFL players that could run a 4:30 mile right now.

These are great athletes and most of them don't run under 4:30 for 1500m.
Hans Van Alphen weighs about 200 pounds and has run a 4:17 1500m, which would be about a 4:36 mile.

I would estimate that there are about 300 athletes under 200 pounds. Here is some data from two years back.
With this many guys I can see why you said there is a chance, but I think you're under estimating the difficulty of a 4:30 mile.
I'm a 230 lb, 34 year old powerlifter who just started running a few years ago. I have a permanently detached hamstring and recently had a dislocated hip, a broken pelvis, and a fractured ankle. I run three times per week.

You just proved my point. You are not a high level athlete, thus you can't comprehend what a high level athlete can do (I am not saying I am either). 4:30 is not that fast. Now if we are talking sub 4:10 or something then yes, I think it requires specific training.
